they are a part of the whole chrome trim. There are two nuts on the trim itself on the inside behind rubber caps. THe rest (chrome bit) to take it off you will need to take off your entire door trim top and bottom and undo some 15 nuts that hold it on, Its a bit of a bitch as the window wont wind down enough for the bottom nuts. Also the mirror must come off.
When you remove the door trim you will work it out quite easily.

Kanchana, the B-pillar covers aren't hard to take off. They are only held on by double-sided tape. I had ours redone (in a dark gunmetal grey) not long after we got the car. If you try lifting them off you can see the tape- just slice and pull gently. I used new tape and some sticky black mastic to retain them, a year later no problems.
From memory there are two screws in there but they don't help to keep the cover on.

The black caps hide two screws which from memory go into the backing for the B-pillar (or rather, the B-pillar itself) and don't have anything to do with holding the cover on.
Try taking them out, it won't hurt, but I recall looking at the inside of the screws after taking the cover off (i.e. the thread sticking through the B-pillar).
Be sure to get the really good "outside exposure" double-sided tape as that's pretty much all that's holding the covers on, unless you put a few globs of panel-beater mastic on too as I did.
They are not easy to get off, Toyota uses really good tape, you need to lever up from the side (protecting door paintwork in the process) and then stick a very sharp blade in and make gentle cuts in the tape- it will get easier after the first side is lifted. The door function isn't altered and you can drive around with the covers off- they are cosmetic.
